3个地理居群泥蚶核糖体DNA ITS区的RFLP分析

利用PCR-RFLP方法对3个地理居群泥蚶的核糖体DNA ITS区进行分析,从分子水平探讨三者的遗传多样性及亲缘关系.结果表明:在选用的6种限制性内切酶中,BstHH I、Hae III和Sse9 I为多态性的限制性内切酶,Msp I、Rsa I和Taq I为单态性限制性内切酶.对酶切图谱进行数理统计后发现宁波海区泥蚶群体内核苷酸多样性指数π值为0.001 7,温州海区泥蚶π值为0.001 6,而莆田海区泥蚶π值为0.001 1,说明宁波海区泥蚶群体内遗传多样性最大,温州海区群体内遗传多样性次之,莆田海区群体内遗传多样性最小.对所有酶切图谱的复合单倍型进行统计,共发现10种复合单倍型,其中宁波... 2RFLP analysis was used to investigate the genetic diversity and phylogenetic relationship of three populations of Tegillarca granosa based on the ribosomal internal transcribed spacer (ITS) region. The results showed that BstHH I, Hae III and Sse9 I were polymorphic restriction endonucleases in PCR product digestion, while Msp I, Rsa I and Taq I were monomorphic. Nucleotide diversity index (π value) of population in Ningbo was 0.001 7, and π values in Wenzhou and Fujian were 0.001 2 and 0.001 1, respectively. It was shown that the genetic diversity was the highest in Ningbo population followed by Wenzhou population and Ningbo population. According to the Pnet value among three populations and cluster analysis results, it was concluded that the phylogenetic relationship between Fujian population and Wenzhou population was farther than that of Fujian population and Ningbo population and the geography relationship of Wenzhou population and Ningbo population was the closest.
