520 |a In a perfusion model using isolated small intestine of the pig, a strong inflammatory reaction with cytokine release and induction of corresponding mRNA expression was shown during reperfusion with whole blood. This reaction already occurred during cold ischemia time (CIT), being far more pronounced after 20h CIT than after 2h CIT. Reperfusion can cause mucosal damage, whereby a prolonged CIT leads to an earlier and initially greater mucosal damage after reperfusion. As an expression of the inflammatory reaction mediated by ischemia-reperfusion injury (IRI), we observed an increased expression of IL-6, IL-2, HSP70, IFN-gamma, and E-selectin. Using in-situ-hybridization, we were able to detect the mRNA expression of IL-6 not only in immuncompetent cells but also in endothelial, smooth muscle, and ganglia cells. Therefore it may be deducted that these cells also, besides the immuncompetent cells, play a considerable role in the initiation of intestinal IRI
