An approach often used for the discovery of new lead structures is the screening of large sets of compounds for inhibitors. Therefore strong research activity has been focused on the automation and optimisation of the screening of libraries with high troughput (high-throughput screening, HTS). The most demanding prerequisite for this kind of strategy for the development of inhibitors as well as for basic biochemical characterisation of the enzyme family was an adequate, non-isotopic, biochemical assay for HDAC
